In vitro Culture and Morphometry of Porcine Adipose Derived Mesenchymal Stem Cells (pAD-MSCs)

Background: Mesenchymal stem cells are well known for their self-renewal capacity and ability to differentiate into multiple cell lineages. The aim of the study was develop a simple technique isolation mesenchymal from porcine adipose tissue morphometric characteristics cells. Methods: Porcine derived were isolated in vitro by using collagenase type II enzyme. Cell yield viability calculated trypan blue exclusion method Neubauer’s chamber. Characterization MSCs done specific markers. morphological changes, morphometry analysed culture Leishman’s stain. doubling (CD) Population time (PDT) also calculated. Result: adherent start forming colony demonstrated an elongated, round spindle like fibroblastic morphology day 1. Almost 80-90 per cent confluency attained on 8-9 after initial seeding reduced 3-4 subsequent passages. RT-PCR reactions revealed positive expression markers CD44, CD73 negative CD34, hematopoietic surface marker. Immunocytochemistry CD44 CD34. In studies, length, nucleus width increased between 24 48 hours both P2 P3.
